Friend is trying to use his Amazon fire stick upstairs in his house, but has an error message of 'Weak or no signal', he has pretty good fibre broadband and the hub is in his living room pretty much directly below downstairs, haven't had chance to get around to have a look yet but can any Fire Stick users on here shed any light, would it be something to change in it's settings or would he need a wi-fi booster of some sort and if so, any suggestions?

What hub? BT? You may need to split the 2.4 & 5Ghz Wi-fi channels, figure out which is the best and connect the firestick to that one.

I had this issue with a Chromecast, and fixed it by buying:

https://www.amazon.co.uk/gp/product/B01BECPIMC/ref=od_aui_detailpages01?ie=UTF8&psc=1

and

https://www.amazon.co.uk/Ugreen-Ethernet-adaptor-Chromecast-network/dp/B01N1X28F1

Basically the first one makes your mains circuit capable of transmitting data (one plug connected by the router, the other wherever he wants to receive the data) and the second link enables you to plug the chromecast (and hopefully firestick - check it's compatible??) into this via an ethernet cable - so essentially a wired connection direct from the router to the firestick via your mains circuit.

This is a possible issue, the router will transmit outwards, but the amount of signal it can send vertically is obviously limited by the design of the hub and how it’s antenna are configured. There are ways you can tweak it, by putting aluminium foil at the back of the router and altering the shape to see how it affects the signal, it’s acting as a reflector. I used to do it with mine, taping cooking foil to the back, and moving the router around to get a better signal in my bedroom, above and to one side of the room with the router. I’ve now got the latest generation of HomeHub, and it seems to be much better at getting a signal around the house, and out the back of the house as well, but that’s a bit more hit-and-miss, but in what is now my bedroom, directly above the router, I get a consistently good signal showing on my phone.

I think there is also a recommendation to use am extension cable to move the firestick from the back of the TV to the side, so the TV itself isn't blocking the signal. Might be worth a try for a few quid.

I'd just buy one of those plug in wifi repeaters and either plug it in in the offending room or landing (whichever works best).

They can be had for around £15 job jobbed.
