I was furloughed for 3 weeks at the start of the pandemic but my employer has told me they paid me too much for that month.
They now want the excess back in this month's pay.
Can they take it all at once? It's around £250 😤
Yes they can providing it's actually a mistake as they're not taking it back; they've effectively advanced it to you. Only if they'd done it for a number of months would it be unreasonable to ask for it back in one go.

When t was discovered I was overpaid over a long period of time total came to £800 ish, they wanted it all back next pay date. I argued that I was overpaid by small amounts stake it back in small amounts as taking the full amount off in one go would leave me with almost no pay.

The answer is yes but only if they don’t put you into hardship. I was overpaid a few years ago as they laid me overtime twice, I informed them as soon as opens my payslip. It was a significant amount so I offered to BACS straight back. After a month they finally decided to take it off me over 6 months, i insisted on paying them back in one. As they refused I just seen it’s a free loan.

It would be not taking holiday and keeping the pay, which is the same as having worked x extra days and hours for free.
