I have quite a few drill holes etc on the external wall of my house (they were there when we bought it last year) and I'm looking to fill them in before the winter.

Is cement best? Or is there a fancy glue or something that's better?

Yes cement based mortar is best for the job. Mortar is the mix of sand and cement used to build brick walls. You can buy a ready mixed tub of mortar from the usual DIY places, just add water.

If it is in brick you can drill another similar coloured brick (or hidden brick) and push the dust onto the surface to hide the cement colour. Can use dye too but the dust alone can work wonders.

If its in painted render rather than brick, Toupret exterior filer will do the job.

I'm also looking for a bit of advice, on how best to fill a hole!
Had a 4 inch hole in a roughcast cavity wall for a tumble dryer duct.
I moved the tumble dryer and cored a new hole, I still have this core and the one for the internal wall.
How best to make it look half decent?

Either silicon the core in the hole and make good or just leave the vent in place.
