Long story short, my partner is a US Citizen and will be flying over to England on the 3rd September and staying with me until we both scarper off to Greece on the 5th September.
Does anyone know or can anyone point me in the right direction regarding isolating, booking tests etc. etc? I have looked on the gov.uk website to no avail and my Google-fu is definitely lacking when searching for an answer?
If it helps, we return from Greece on the 17th September then she returns to the US on the 25th September.
Thank you all.

My reading on that is that on both occasions she will have to treat the 'layover' as a proper entry to the country and follow the rules as if she were visiting the UK, because she is coming landside and not travelling directly to the port of exit.
Someone else may be able to help her vis a vis obtaining tests.
Good luck!
I think the issue is this bit
---
On arrival in England
On arrival in England you must:
quarantine at home or in the place you are staying for 10 days
take a COVID-19 test on or before day 2 and on or after day 8
---
The rules do not seem to allow a couple of days here and there - you must quarantine for 10 days.
Given you and your partner's proposed plans I would check careful you'll not have any issues with your onward travel to Greece.
My sister did a 3 day visit last month under these rules - she just complied with the bits she cold then flew home again on day 4. No issues leaving the country
IMO i'll be suprised if UK citizens will be able to fly anywhere shortly..
Plague Island will be red listed from nearly everywhere.
If we are still allowed to travel it might be worth her flying straight to Greece.
martinhutch has it. The law does not require you to stay for 10 days, you can leave the UK sooner. So she needs to do the pre departure test, plf and book the day 2 and 8 test. Because she'll have left the uk again she may be able to use the leftover day 8 test posted to your house to subsequently leave the UK back to the US if she wanted to, although rapid tests also suffice for the US.
Do note the CDC have literally just moved the UK to Level 4 so she will want to check her travel insurance to know what she is covered for, etc.
