I'm PAYE and do part time (very) ad hoc work. I drive my car to wherever I'm working in the UK and claim a mileage allowance of 28p per mile. Can I also claim VAT on top of that?

No, because you aren't VAT registered. What you can do is claim the tax relief on the difference between 28p and 45p for the first 10,000 miles.
