As it says can I leave the cassette on?

Can't see why not if a) it doesn't interfere with getting it in your jig and b) you haven't got one of those wheels with the nipples at the hub end of the spokes.

What cassette is it? If it's Dead or Alive's You Spin Me Round - then yes.

over tighten and accidentally break a spoke, then best to have Cassette off to begin with... 😉

Yes, you'd have to remove the cassette if its on the drive side. So you take it off just like any other wheel with a broken spoke.
