Depending on the rear hub, you might be able to rearrange the spacers, to achieve the correct spacing.

In that case, you'll be no worse off than if you were on the granny or big ring.
Out of interest, why aren't you running it in the position it was designed for?
It is the only front ring that you're running, isn't it?
I would think it will be fine. You should be able to get the full cassette spread with the middle ring on a 3 ring set up.
If you feel the chainline could be better and you have a spacer on the none-drive side of the BB then you could always shift it over to the drive side. Just check heal-chainstay clearance if it's already close.
