Not had this quite before.

One gear in the middle of an 11spd block (4th smallest) can't be made to run smoothly. Ultegra 11spd mechanical. Gears above and below all tip top. The drive train has only done circa 300 miles but not bedding in and no amount of fettling cable tension helps. Not enough to make the chain skip gears - it's just snickerty. Can't see a manufacture error. It's a miche cassette so easy enough to swap out I guess.

Frustrating.

I've not had issues with them before.......I guess first time for everything.

I've had the same on an XT 11 speed setup.
All ran ok and shifted fine but on one cog it would just catch and rub a bit, not enough to make the gear skip or anything but really annoying.
Tried all sorts including gear hanger allingnment which made it worse when it was spot on.
In the end I just experimented with bending the mech hanger a tiny bit at a time and tuned it out fine.
I did this by putting an allen key in the mech hanger bolt and tweaking it like that so as not to bend the mech its self.
Tweek, ride, tweek, ride, tweek, perfect.

Had this with wife’s cassette, turned out I’d put the cog on back to front by accident, might be worth checking if the one in question is a separate piece

Just binned a SRAM Eagle cassette as the chain refused to change down from one cog. Even on close inspection, I really couldn’t see what the problem was with the cassette, not particularly worn, chain is fine, mech straight and the derailleur‘s pretty new. I decided to try a new cassette and problem solved.

Was thinking cog in back to front or possibly spacer on wrong side of the cog.

bent mech / mech hanger.
I'd say sticky cable but if it's new it's unlikely. It's always been one of those 2 when I have had issues in the middle cogs rather than the ends. Just replaced a rear mech recently for this, which had been bent and I'd worked it back myself but it's never been quite right.
