With the guard in the correct position not to foul at full suspension compression.
Fitted mine and the wheel spins freely, but there isn't a huge clearance at the top of the tyre.

About 7mm on my five but I did shave it a bit more for extra clearance.

I've got about 5mm with HR 2 2.3 tyres on.
Might shave a bit off as it gets clogged with mud quite easily.
It's a good rear guard though.

Got mine running at 10mm on my orange five, with a specialized storm control 2.0

I left a fingers width gap between mine and the wheel, which gives good enough clearance for me (Nukeproof Mega/Specialized Butcher).

With a 2.4" Schwalbe Fat Albert in the back of my Guapo, the tyre rubs where the guard straights to straighten out.
I've filed the top of the guard to make it fit 'into' the seattube under full compression a bit more but not ideal with big tyres. A 2.25" maxxis seems fine. A 2.0" Spesh storm in now is much better

Does the standard rear Mudhugger fit over a 29er tyre... I have seen them on 29ers but not sure if they are a specific 29er guard.

Can't see a 29er one on the website but would like to be sure.

Cheers
