Any suggestions for a proper (50k+) 3 days of riding (back to back) around the South of England or Wales?

The south downs way is a good one. Picturesque, but not properly wild. 160k so fits your 3 day schedule well.
King Alfred’s way (or parts there of according to distance) has some interesting history and very varied landscapes.

Forgot to mention that we'll be on a campervan and everyone is riding... so loops or 'out and back' is essential, rather than point to point.

Lots of big loops available starting from the Elan Valley.
There are lots of loops you can do along the SDW, taking in some of the main route and adding up and down to the N and S of the main ridge
