I have a 10spd SLX rear mech for sale that nobody seems keen on buying so I think I'll just stick it on the Mrs' bike.
Can I run 1 x 10 with a long cage rear mech? Or should I stick to 2/3 x 10?

Should be okay. Shorter will keep more tension which is better but not necessary. Though if your long cage is a clutch mech then that will make up for it anyway.
Make sure the chainring is a narrow-wide.
