Aero road frames..do they actually make a difference?

More i read about aero frames the more intrigued I am. Currently riding a canyon ultimate with and it's not a great deal faster on the flat than my allez winter roadie. But stick on my 50 mm carbon wheels and the average speed of my rides jumps up by up to a mph.

All the manufacturers tell us aero is far more important than weight, and although I like a light bike and enjoy climbing I would like a bit more free speed to help me on the flats where I struggle to keep up with my more powerful training partner (my power to weight is respectable and i can keep up on the climbs no problem)

SO will I instantly gain a mph by buying something like a propel, or in reality is the difference all in the wheels. Already got an aero helmet, so that cheap upgrade is already taken care of. Position on the bike on the canyon is already pretty low, so the difference would be purely in the shape of the frame tubes, rather than a lose front end.
