OK, so a Dyna sis 10sp mech needs 10sp Dyna sis shifter (or does it? any good ways to get a 10sp thumbshifter for this?), but which cassette does it have to be - can any 10sp shimano cassette be used? And what about SRAM?

yeah, as long as it has 10 sprockets, it will be fine. Brand is irrelevant for what cassette to use.

Microshift make a 10 speed thumbshifter. Paul components makes a mount that lets you use Shimano Dura Ace bar end shifters as thumbies.
